HuProt™人全蛋白质组芯片
HuProt™ HuProtTM V4.0芯片上有超过2万个酿酒酵母表达, 带有GST和His标签的重组人蛋白和部分小鼠重要蛋白, 其中89%为全长蛋白,涉及约17000个人类基因和124种鼠对照蛋白。酵母蛋白具有翻译后修饰, 故也适用于PTM研究。非接触喷印 (Arrayjet Non-Contact Piezoelectric Print) 保证了样品点均匀度和复点间一致性, 分析结果更准确。
检测时,分析物(Analyte)先与芯片孵育, 并与特定蛋白结合。分析物是血液IgG或杂交瘤培养上清时, 使用荧光标记的抗人(鼠)IgG抗体即可检出结合的IgG, 对其他分子则通过标记分析物或其抗体的方式检测,荧光扫描芯片取得数据后用于分析, 后续针对初筛得到的蛋白定制小规模芯片进行大样本验证。
